Biosynthesis of Protein Amino Acids in Plant Tissue Culture IV Isotope Competition Experiments using Glucose-U-C and Potential Intermediates.

چکیده
The isotope competition method with glucose-U-(14)C as a carbon source has been used to determine whether or not selected compounds contribute carbon to the biosynthesis of protein amino acids in cells derived from Paul's Scarlet Rose. Of 48 compounds tested, 15 contributed carbon to protein amino acids. The results show for the first time that homoserine is an intermediate in threonine biosynthesis; that homoserine, cystathionine and homocysteine are intermediates in methionine biosynthesis and that histidinol is an intermediate in histidine biosynthesis in plant cells.

Biosynthesis of Protein Amino Acids in Plant Tissue Culture II Further Isotope Competition Experiments Using Protein Amino Acids.
The behavior of exogenously supplied alanine, proline, glutamine, glutamic acid and aspartic acid in isotope competition experiments using cells of Paul's Scarlet Rose grown in the presence of glucose-U-(14)C was examined at higher concentrations than were used in part I of this series. Each amino acid contributes significantly to protein above a minimum concentration in the culture medium. Ther...
متن کاملThe biosynthesis of protein amino acids in plant tissue culture I. Isotope competition experiments using glucose-U-C14 and the protein amino acids.
Most investigators have used excised tissue, cellfree preparations, or whole plants for studies of metabolism in angiosperms. Each of these systems represents a different state of the cells or tissues. Another state of plant cells, that of exponential growth, has been obtained by tissue culture techniques (2, 5). Studies of the metabolism of cells growing exponentially would provide information...
متن کاملIsotope labelling of Rubisco subunits provides in vivo information on subcellular biosynthesis and exchange of amino acids between compartments
The architecture of plant metabolism includes substantial duplication of metabolite pools and enzyme catalyzed reactions in different subcellular compartments. This poses challenges for understanding the regulation of metabolism particularly in primary metabolism and amino acid biosynthesis. To explore the extent to which amino acids are made in single compartments and to gain insight into the ...
متن کاملDynamic Labeling Reveals Temporal Changes in Carbon Re-Allocation within the Central Metabolism of Developing Apple Fruit
In recent years, the application of isotopically labeled substrates has received extensive attention in plant physiology. Measuring the propagation of the label through metabolic networks may provide information on carbon allocation in sink fruit during fruit development. In this research, gas chromatography coupled to mass spectrometry based metabolite profiling was used to characterize the ch...
متن کاملDiminished Superoxide Generation Is Associated With Respiratory Chain Dysfunction and Changes in the Mitochondrial Proteome of Sensory Neurons From Diabetic Rats
OBJECTIVE Impairments in mitochondrial function have been proposed to play a role in the etiology of diabetic sensory neuropathy. We tested the hypothesis that mitochondrial dysfunction in axons of sensory neurons in type 1 diabetes is due to abnormal activity of the respiratory chain and an altered mitochondrial proteome. RESEARCH DESIGN AND METHODS Proteomic analysis using stable isotope la...
